SRX25680668: WGS of Leishmania mexiacana TERT_KU80_mut
1 OXFORD_NANOPORE (PromethION) run: 229,491 spots, 3.1G bases, 2.7Gb downloads

Design: Genomic DNA (gDNA) from 2 10^8 L. mexicana cells was isolated using the phenol/chloroform method. DNA integrity was confirmed via 1% agarose gel electrophoresis. Samples were purified with the Short Read Eliminator XS kit (PacBio) and quantified using a Qubit 4 Fluorometer (Thermo Fisher Scientific). Approximately 1 g purified gDNA was used for nanopore sequencing library construction with the Native Barcoding Kit 24 V14 SQK-NBD114.24 (Oxford Nanopore Technologies) and sequenced on a PromethION Flow Cell.

Study: Ablation of LmKu80 activates alternative lengthening of telomeres in Leishmania mexicana

The study investigates the impact on telomere maintenance in L. mexicana through knockout mutants of Ku80 and TERT, as well as a double-knockout of both genes. To provide insights into the behavior of telomeres we employed long-read nanopore sequencing for the analysis of chromosome arm-specific telomere reads.
